On returning to India, she met noted singer Kumar Sanu in Kolkata. On his recommendation, she then made her playback singing debut through Rajesh Roshan's Moksha. She has then gone on to sing songs in films like Yuva, Aayitha Ezhuthu, Kal Ho Naa Ho, Anbe Aaruyire and Kuch Naa Kaho, and her most recent albums include A.R. Rahman's Guru, and Rang De Basanti, Srikanth Deva's Aalwar, Yuvan Shankar Raja's Deepavali and Sarvam and Harris Jayaraj's Unnale Unnale. Her song Vaaji Vaaji in A.R.Rahman's Sivaji was well received.

She sang 3 songs in A.R.Rahman's Tehzeeb, for which she has been credited as Sujata Bhattacharya. Though these songs did not become popular, they are considered by critics, to be among the best songs sung by her[citation needed. But the song she is known for most is "boondon se baatein" from thakshak. She also sung a song 'In Lamhon Ke Daaman Mein' from the movie Jodhaa Akbar, music directed by A. R. Rahman.

Originally hailing from a Bengali background, Madhushree sang Tamil and Telegu for many South Indian composers. Her voice became an instant hit, especially under the discography of A. R. Rahman. Finally, Madhushree learned Tamil on her own wits to grasp a better understanding of her work and passion of singing, but still, her diction in Tamil is not perfect, according to critics[cite this quote. Currently married to engineer and music director Robby Badal, Madhushree will next be seen singing for Karthik Raja's Mukhbir.
